What about the "favourite" option in the twitter interface? On http://twitter.com it's the little star next to the tweets, below the "reply" icon, and in most API front-ends it's either a heart (like in twhirl) or an un-icon'd contextual menu item (like in tweet deck). TweetDeck even has a way to display just favourites and then add/remove them as you like! :)
In fact, I definitely recommend you check out http://tweetdeck.com; it's the one everyone seems to be using if they follow more than they can keep up with, as it allows groupings and live hashtag-searchings and filterings and various other lovely and productive things :)
